摘要:首先采用Box-Behnken试验对纤维素酶提取双孢菇多糖的工艺参数进行优化,然后研究其抗氧化活性,最后分别将其与其他提取液调配成复合饮料。实验结果表明:最优纤维素酶提取双孢菇多糖的工艺参数为酶浓度0.72%、pH值4.3和酶解时间108min,此时得率为10.55%;当多糖浓度在16.32μg/mL~163.2μg/mL和13.06μg/mL~104.45μg/mL的范围内,其清除DPPH自由基和羟基自由基的能力分别为35.79%~76.54%和23.44%~83.71%,且清除效果与浓度之间都存在明显的量效关系;双孢菇分别和芦荟(桂花、桑白皮、茉莉花、菊花、金银花)复合饮料的最佳工艺参数皆为配比1:2、提取液所占饮料比1:10、白砂糖添加量10%和柠檬酸添加量0.10%,此时双孢菇复合饮料的口味最佳。

Abstract:First, the technolgical parameters of cellulase extraction of agaricus bisporus polysaccharides were optimized basing on the Box-Behnken experimental design, then the antioxidant activity of agaricus bisporus polysaccharides were also be researched and the last the compound beverage was mixed with agaricus bisporus polysaccharides and other extracting solutions. The results show that the best optimum extraction conditions were obtained as follows: enzyme concentration 0.72%, pH value 4.3 and enzymolysis time 108 minutes, under the extraction conditions, it can be included that the extraction yield is 10.55%; When the concentration of the polysaccharide is 16.32μg/mL~163.2μg/mL and 13.06μg/mL~104.45μg/mL, the clearance rate of DPPH and ·OH free radical are 35.79%~76.54% and 23.44%~83.71% individually, and the scavenging activity is correlated with the concentration of the polysaccharide. The best technolg- ical parameters of compound beverage of agaricus bisporus and aloe vera(osmanthus, white mulberry, jasmine, chrysanthemum, honeysuckle) were agaricus bisporus sample solution to aloe vera sample solution 1:2, drink mixture 1:10, the adding amount of sugar 10% and amount of citric acid 0.10%, under these conditions, it tastes best.
